摘要 目的 :观察 C2 -神经酰胺诱导肝癌细胞凋亡 ,为进一步研究肝癌细胞凋亡信号的传导提供实验依据。 方法 :将 C2 -神经酰胺作用于肝癌细胞 SMMC- 772 1及 BEL- 740 2后 ,分别行 H- E染色、荧光染色、TU NEL 方法检测及流式细胞仪 Sub- G1法 ,检查细胞的凋亡。结果 :10 μm ol/ L C2 -神经酰胺作用肝癌 SMMC- 772 1细胞 ,48h后发生凋亡 ,H- E染色及荧光染色检测可见典型的细胞凋亡表现 ,细胞核固缩、碎裂 ,以及碎裂的细胞核浓染成为凋亡小体。经流式细胞仪检测可见典型的 Sub- G1凋亡峰 ,凋亡细胞占细胞总数 (9.2 0± 0 .73) %。在 BEL- 740 2细胞也观察到相似的结果。 结论 :C2 -神经酰胺可诱导肝癌SMMC- 772 1及 BEL- 740 2细胞的凋亡。 Objective: To observe the apoptosis of hepatoma cells induced by C 2-ceramide, providing experimental data for further research on the signal transduction of apoptosis in hepatoma cells. Methods: Hepatoma cells, SMMC-7721 and BEL-7402, were detected with H-E staining, fluorescent staining, TUNEL staining and flow cytometry after incubated with 10 μmol/L C 2-ceramide for 48 h. Results: In SMMC-7721 cells, the apoptotic body and condensation of chromatin were detected with H-E staining and fluorescent staining. The apoptotic hepatoma cells were also detected with the TUNEL method. The apoptotic peak were seen by the DNA content assay with flow cytometer and the apoptotic hepatoma cells accounted for (9.20±0.73)% of the total SMMC-7721 cells. Similar results were seen in BEL-7402 cells with H-E staining and fluorescent staining method. Conclusion: C 2-ceramide can successfully induce apoptosis in SMMC-7721 and BEL-7402 hepatoma cells.
