摘要
目的 了解第二信使神经酰胺信号转导对鼻咽癌细胞生长的影响。方法 采用MTT法检测神经酰胺对鼻咽癌细胞株 (CNE 2 )增殖的抑制作用。光镜观察、荧光染色、流式细胞术检测细胞凋亡改变。Westernblot法检测p2 1WAF1的表达。结果 神经酰胺在 6 2 5、12 5、2 5、5 0 μm浓度下 ,对CNE 2细胞生长有明显的抑制作用 ,流式细胞仪检测发现亚G1峰出现 ,荧光染色可见核荧光强度增加、核片段化和凋亡小体。p2 1WAF1蛋白表达明显上调。结论 神经酰胺能诱导鼻咽癌细胞株CNE 2凋亡 ,且上调 p2 1WAF1蛋白表达。p2 1WAF1可能是神经酰胺诱导鼻咽癌细胞凋亡中起作用的因素之一。
AIM To investigate the induction of apoptosis of nasopharyngeal carcinoma cell line CNE 2 by ceramide signaling METHOD MTT assay, microscope, fluorenscein staining, flow cytometry(FCM) and Western blot were used RESULTS CNE 2 treated with C 2 ceramide underwent cell proliferation inhibition IC 50 value was 16 25 μmol·L -1 ; cell shrinkage, condensation cell chromatin clinged to periphery of the nuclear and fragment were detected by fluorenscein microscope Apoptosis peaks were also detected on FCM p21 WAF1 expression in CNE 2 treated with ceramide was observed by Western blot CONCLUSION Ceramide can induce apoptosis of CNE 2 cell line p21 WAF1 expression was upregulated in CNE 2 treated with C 2 ceramide
