一氧化氮和抗氧化酶活性与动脉粥样硬化关系的研究 被引量:7

摘要 目的 :从整体及细胞水平上对一氧化氮 (NO)、抗氧化酶活性与动脉粥样硬化的关系作一初步探讨 ,同时观察阿魏酸钠 (sodiumferulate ,SF)对其的保护作用。方法 :以实验性动脉粥样硬化 (atherosclerosis ,AS)鹌鹑做为动物模型 ,动态观察其血中NO的生成和抗氧化酶活性的变化以及SF的影响 ;以高脂血清培养的人脐静脉内皮细胞作为细胞模型 ,观察其脂质过氧化水平、NO产生的情况以及SF的保护作用 ;以氧化低密度脂蛋白 (OX LDL)抑制巨噬细胞NO的产生 ,同时观察SF对其抑制作用的影响。结果 :①高胆固醇血症可显著升高血浆脂质过氧化物(LPO)水平 ,降低血浆NO水平和显著降低红细胞超氧化物歧化酶 (SOD)、谷胱甘肽过氧化物酶 (GSH px)比活性的作用。②SF有清除自由基、减轻膜脂质过氧化、提高抗氧化酶活性的作用。③高脂血清 (HRS)可使培养的人脐静脉内皮细胞中LPO含量升高 ,并随着HRS浓度的增加 ,LPO含量逐渐升高 ;VitE可阻止HRS引起的内皮细胞中LPO含量的增加。④SF具有与抗氧化剂VitE相似的保护内皮细胞释放NO的作用。⑤OX LDL对脂多糖 (LPS)诱导的巨噬细胞NO的产生具有抑制作用。⑥SF对OX LDL抑制巨噬细胞NO的产生具有明显的保护作用。结论 :提示SF在保护血管内皮细胞。 Objective: To study the relationship of nitric oxide(NO),activitives of antioxidase and atherosclerosis(AS) from the levels of whole body and cell,and to study the protective effect of sodium ferulate(SF). Methods: The animal model is the experimental arteriosclerotic quail, dynamic observation the production of NO and activitives of antioxidase in blood and the protective effect of SF on it; the cell model is the cultured human vascular endothelial cells(EC) on hyperlipidemic serum (HRS), observation the level of lipid peroxidation(LPO),the producation of NO and the protective effect of SF on it; inhibition the producation of NO in macrophges by oxidized low disity lipoproteins(OX LDL),and observed the effect of SF on it. Results: (1)Hypercholesterolemia can raised the LPO levels in plasma, and decreased the NO production in plasma,and decresed the specific activity of SOD and GSH px in red cells markedness. (2)SF has the effects of scavenging free radical and abatementing the lipid peroxidament of cell membrane and increasing the activity of antioxidase. (3)HRS can increase the LPO in cultured EC and Vit E can inhibatant the increasing of LPO in EC. (4) SF has the same effect of protection the producing of NO in EC as Vit E. (5) OX LDL can decrease the NO production in macrophges induced by LPS. (6) SF has the protective effect on NO production of macrophges. Conclusion: SF has apparent effect on protective EC and prevent AS.
