摘要
本文主要研究了采用响应 pH -stat的流加培养模式 ,进行Xanthophyllomycesdendrorhous高细胞密度培养及虾青素合成。在该补料间歇培养模式中 ,发酵过程 pH的恒定控制采用 2 .78mol L葡萄糖及 3mol L氨水来控制 ,同时起到流加葡萄糖的作用。试验结果表明 ,虾青素积累量于 78h可达 4.0 5mg L ,菌体干重在第 84h获最大值 14.6 2g L。与不流加葡萄糖、氨水的间歇培养相比 ,虾青素含量和菌体干重分别增加 2 .89、5 .2
To examine the new approach for high cell density fermentation and high production of astaxanthin in the red yeast of Xanthophyllomyces dendrorhous , fed batch with pH stat controlled by 3mol/L ammonia and 2.78 mol/L glucose was performed, where pH change were responded by glucose and ammonia, not sulfuric acid and sodium hydrate. The experiment results showed that DCW (dry cell weight) was high by 14.62g/L at 84h, and astaxanthin volumetric yield was up to 4.05mg/L at 78h. Compared with the batch fermentation without pH stat, the productivity of astaxanthin and DCW were improved by 2.89 and 5.22 times respectively.
