摘要 目的 探讨 1 -methyl-4 -phenylpyridium iodide( MPP+)对 SHSY5Y细胞的毒性作用。 方法 采用体外培养的 SHSY5Y细胞 ,通过 TUNEL染色、细胞超微结构观察和 DNA倍体分析检测 MPP+对 SHSY5Y细胞凋亡的诱导作用。 结果  MPP+可明显地抑制 SHSY5Y细胞的生长 ,并诱导 SHSY5Y细胞发生凋亡 ,主要表现为TUNEL染色阳性的细胞增多 ,细胞核异染色质增多 ,线粒体肿胀、嵴断裂、消失 ,在 DNA周期中的 G0 -G1期前出现明显的亚二倍体峰。 结论 MPP+通过诱导 SHSY5Y细胞凋亡的方式发挥毒性作用。 Objective\ To study the toxicity of 1\|methyl\|4\|phenylpyridium iodide(MPP\++) on SHSY5Y cells in vitro.\ Methods\ The SHSY5Y cells were used as cell model.\ The apoptosis of SHSY5Y induced by MPP\++ was observed by TNNEL, ultrastructure and flow cytometry.\ Results\ It showed the growth of SHSY5Y was obviously inhibited by MPP\++ and apoptosis was induced.\ There were more TUNEL positive cells in MPP\++ group than control, a marked subdiploid peak before G\-0\|G\-1 on DNA cycle, condensed nuclei under transmission electric microscope, and swelling of mitochondria with break and disappearance of their cristas.\ Conclusion\ The exogenous MPP\++ exhibits its toxicity as inducing apoptosis in SHSY5Y cells.\;
