摘要
目的研究水蛭提取液的抗凝和纤溶作用。方法观察水蛭水煎液及其醇溶和非醇溶部分抑制凝血酶引起纤维蛋白 (Fb)凝块形成的活性 ,纤维蛋白平板和SDS 聚丙烯酰胺凝胶电泳分析各提取液水解Fb和纤维蛋白原 (Fbg)的作用。结果水煎液及其醇溶部分可抑制Fb凝块形成 ,非醇溶部分无此作用。水煎液及其非醇溶部分可水解Fbg和Fb ,且以α链为主 ,对β和 链及牛血清白蛋白的水解作用弱 ;醇溶部分对这些蛋白的水解作用不明显。结论水蛭水煎液的醇溶和非醇溶部分分别含抗凝和纤溶物质 。
PurposeThe aim is to explore in vitro anticoagulant and fibrino(geno)lytic activities of water decoction of leech (LWD).MethodsAnticoagulant activity of LWD and its ethanol soluble (LWDES) and inso luble part (LDWEIS) were determined by the inhibition of thrombin induced fibrin clot formation respectively. Fibrin plate and SDS polyacrylamidegel electrophoresis (SDS PAGE) were used to investigate the hydrolytic activities of the three leech′s extracts against fibrin and fibrinogen.ResultsLWD and LWDES completely in hibited fibrin formation,LWDEIS had no such effect. LWD and LDWEIS degraded fibrinogen and fibrin with preference against α chains rather than β and chains and bovine serum album. No apparent hydrolytic activity of LWDES against any of the substrates was found.ConclusionLWDES and LWDEIS contained anticoagulant and fibrino(geno)lytic substance respectively. The fibrino(geno)lytic activities of LWD and LWDEIS were characterized by relative substrate specificity.
