摘要
本项实验依据中医肾虚 骨质疏松 髓海 (脑 )不足的理论 ,通过建立肾虚骨质疏松症大鼠动物模型 ,应用补肾中药密骨灵实验组和各对照组灌胃治疗 ,检测大鼠下丘脑 5 羟色胺 (5 HT)、5 羟吲哚乙酸 (5 HIAA)、去甲肾上腺素 (NE)、多巴胺 (DA)含量的变化 ,以及通过对大鼠脑总磷脂 (PL)、总胆固醇 (CH)的测定 ,探讨该病证动物模型神经内分泌方面的病理机制 ,阐明补肾中药对该病证神经内分泌病理改变的调节作用机理。结果表明 :肾虚骨质疏松症雌性大鼠脑组织总胆固醇 (CH)、总磷脂 (PL)含量及CH PL的比值明显增高 (P <0 0 1) ;下丘脑NE、DA含量明显降低 (P <0 0 5和P <0 0 1) ,导致 5 HT NE、5 HT DA的比值高于正常对照组 (P <0 0 1) ,提示该病证的病理机制与神经内分泌网络具有密切的相关性。使用中药密骨灵及骨疏康颗粒进行防治 ,取得良好的效果。密骨灵和骨疏康颗粒可使脑组织总磷脂、总胆固醇含量以及总胆固醇 总磷脂的比值有所降低 (P <0 0 1) ,提高NE含量 (P <0 0 5 ) ,降低 5 NT NE、5 HT DA的比值 (P <0 0 1) ,通过延缓脑组织老化 ,改变中枢神经递质水平 。
The experiment is based on the theory of kidney insufficiency\|osteoporosis\|insufficiency of marrow sea(brain) in traditional Chinese medicine.It expounds the pathological mechanism of the neuroendocrine aspects in the osteoporotic biological model and the regulatory mechanism of kidney\|tonifying medicinal herbs on the neuroendocrine pathological change in the osteoporosis.In this experiment,we established a rat model of osteoporosis due to kidney deficiency,gave the kidney\|tonifying herbal medicine Miguling to the experimental group and a definite anti\|osteoporotic herbal medicine Gushukang to the drug control group(the experimental group and other control groups were all administered by gastric gavage) and then examined the content variation of 5\|hydroxy\|tryptamine(5\|HT),hydroxyindoleacetic acid(5\|HIAA),norepinephrine(NE),dopamine(DA) of hypothalamus,and determined the total phospholipid(PL) and total cholesterol(CH) of the rats' brain.The results were as follows:the contents of CH,PL and the ratio of CH/PL in the brain of kidney deficiency\|osteoporosis female rats were higher than those in the normal control model blank groups (P<0.05,P<0.01), and for that reason,the ratios of 5\|HT/NE,5\|HT/DA were higher than those in the normal control group (P<0.01), showing the close correlation of pathological mechanism of this disease and the neuroendocrine network.Both Miguling and Gushukang herbals had good effects.The two herbals can prevent and treat osteoporosis by delaying the aging of brain tissues and altering the levels of central nervous transmitters.
