盗毒蛾核型多角体病毒的分离鉴定 被引量:3

PURIFICATION AND IDENTIFICATION OF PROTHESIA SIMILIS NUCLEAR POLYHEDROSIS VIRUS

摘要 作者从自然罹病死亡的盗毒蛾幼虫中分离到一株昆虫病原物 ,经鉴定 ,该病原物为盗毒蛾核型多角体病毒 (PsNPV) ,该病毒包涵体多数呈三角形、四边形 ,部分呈多角形 ,其大小为 10 0 0~ 2 0 0 0nm ;病毒粒子呈杆状 ,为单粒包埋型 (SEV) ,其大小为 (2 5 0~ 35 0 )nm× (4 0~ 6 0 )nm .通过感染试验测定了盗毒蛾核型多角体病毒的毒力 ,结果显示其LC50 为 3.34× 10 4 PIB/mL ;以 3× 10 6PIB/mL和 3× 10 5PIB/mL两种浓度感染盗毒蛾幼虫 ,测得LT50 分别为 5 .78d和 6 .2 Prothesia similis nuclear polyhedrosis virus (PsNPV) was purified with differential centrifugation on sucrose gradients. It's shape and ultrastructure was studied using electron microscopy .The results showed that the polyhedras were triangle, tetragon or polygon in shape and measured about 1000~2000nm. The dimensions of the virus rods of PsNPV were 250~350nm in length by 40~60nm in diameter. The results of infectivity test showed that the LC 50 of PsNPV was determined as 3.34×10 4 PIB/mL. When the insects were infected at a concentration of 3×10 6 PIB/mL and 3×10 5 PIB/mL, the LT 50 was 5.78d and 6.24d respectively.
