摘要
目的 体外训练单纯疱疹病毒 1型 (HSV- 1)对抗生素 17997的耐药突变及体内、外研究抗生素17997与阿昔洛韦 (ACV)的交叉耐药。方法 HSV- 1在抗生素 17997或 ACV的存在下连续传代 ,选择一定代数测定 HSV- 1野株及训练株的 IC50 。用 HSV- 1的 ACV耐药株感染兔角膜造模 ,分别用抗生素 17997或ACV局部给药治疗 ,以局部病损及病毒分离判断疗效。结果 HSV- 1在抗生素 17997存在下连传 41代仍保持对抗生素 17997的敏感性 ,未产生耐药变株 ;但 HSV- 1在 ACV存在下 ,仅传一代 ,就发生耐药变株。抗生素17997与 ACV无交叉耐药。抗生素 17997对 HSV- 1ACV耐药株实验感染角膜炎有明显治疗效果 ,可减轻病损 ,降低病毒排出量 ;阳性对照药 ACV无任何疗效。结论 体外 HSV- 1对抗生素 17997不易产生耐药。抗生素 17997与 ACV体内。
Object: The resistance and cross resistance of HSV 1 to antiviral antibiotic 17997 were studied in vitro and in vivo . Method: HSV 1 was successively passed in the existence of 17997 and acyclovir (ACV) in VERO cells respectively. The efficacy of 17997 in treatment of ACV resistant HSV 1 induced rabbit keratitis was evaluated by lesion score and virus isolation. Results: HSV 1 was successively passed in the existence of 17997 for 41 generations. There was no any change in the sensitivity of HSV 1 to 17997 with similar IC 50 values between the parent HSV 1 strains and trained HSV 1 strains. On the contrary the resistance of HSV 1 to ACV was very easy to induced. The efficacy of 17997 in treatment of ACV resistant HSV 1 induced rabbit keratitis was the same as 17997 in treatment of ACV sensitive HSV 1 induced rabbit keratitis. ACV did not show any treatment efficacy to ACV resistant HSV 1 induced rabbit keratitis. This means there is no any cross resistance between 17997 and ACV. Conclusion: The results indicated that HSV 1 is difficult to become resistant to 17997. No cross resistance between 17997 and ACV was seen both in vitro and in vivo .
