摘要
目的 :研究抗病毒抗生素 17997与临床应用的抗病毒药物—阿昔洛韦、病毒唑及人干扰素α的联合用药抗单纯疱疹病毒 型的作用。方法 :应用 VERO细胞培养法及 MTT染色法进行实验及观察结果 ,以 Calcusyn软件计算 ,分析结果。结果 :实验结果显示抗生素 17997与阿昔洛韦、病毒唑及人干扰素 α在ED50 、ED70 (或 ED75)、ED90 条件下均有协同抗单纯疱疹病毒 型的作用 ,表现在降低 ED50 值及联合指数 <1。仅抗生素 17997与人干扰素 α联合组 ,在 ED90 条件下 ,表现为相加或拮抗作用。结论 :抗生素 17997抗疱疹病毒活性强 ,在多次单纯疱疹病毒 型实验感染的兔角膜炎模型上显示抗病毒疗效。
Object: To investigate the antiviral activity of antibiotic 17997 in combination with antiviral drugs acyclovir (ACV), ribavirin (RBV) and human interferon α (IFN α) in vitro . Method: Vero cell culture and MTT method were used in the study. The results were analyzed by Calcusyn software. Results: Results displayed synergetic anti HSV 1 effects of 17997 in combination with ACV, RBV and IFN α under the conditions of ED 50 , ED 70 (or ED 75 ) and ED 90 respectively with reduced ED 50 values and combination indexes (CI) less than 1. Only the combination of 17997 and IFN α under the condition of ED 90 displayed an additive or mild antagonistic effect against HSV 1. Conclusion: Antibiotic 17997 is a potent anti HSV antibiotic and shows therapeutic efficacy against HSV 1 inducing experimental rabbit keratitis in vivo . Results showed that antibiotics 17997 could be used as a combined medication in treatment of HSV 1 keratitis and other viral diseases.
