摘要
目的:探讨运用高强度聚焦超声治疗系统(HIFU)制备小鼠肝癌H22固化瘤苗的方法,并初步评价瘤苗的免疫活性。方法:应用HIFU治疗系统样机分别在声强246W/cm2和1054W/cm2下处理小鼠肝癌H22细胞。结果:声强1054W/cm2下细胞破裂成碎片状,而声强246W/cm2细胞外形基本如常,前者作为瘤苗具有明显强的抗肿瘤细胞攻击作用P<0.05。结论:HIFU能一次性灭活肿瘤细胞,并在一定声强下破坏肿瘤细胞膜结构使细胞内抗原充分暴露和释放,从而增加了瘤苗免疫原性。
Objective:To discuss the method of using High Intensity Focused Ultrasound (HIFU) System for tumor therapy to study and develop a solidification vaccine against mouse hepatic cancer H22 cell and to make elementary evaluation of the vaccine immunocompetence.Methods:A Sample machine of HIFU System for tumor therapy with its sound intensity set as 246 W/cm2 and 1054W/cm2 was adopted to deal with mouse hepatic cancer H22 cell. Results: Under the sound intensity of 1054 W/cm2 ,cells were observed to fall into pieces while the cells remained the normal shape under the sound intensity of 246W/cm2 . It was discovered that the vaccine in the former situation was strongly effective in resisting the attack from tumor cells, with P<0.05 .Conclusion: HIFU can destroy tumor cells all at once, destroy cell membrane structure under certain sound intensity and make the complete explosion and release of intracellular antigen so as to enhance the immunogenicity of vaccine.
