雷公藤内脂醇联合热疗对Hep-2细胞的增殖抑制作用 被引量:1

摘要 目的:探讨雷公藤内脂醇(TL)联合热疗对导人喉癌细胞株(Hep-2)的增殖抑制率、细胞凋亡率、及细胞周期增殖各时相的影响,旨在为人喉癌治疗提供理论依据。方法:对数生长期Hep-2细胞随机分为对照组、单纯热疗组、单纯TL组及TL联合热疗组,应用MTT(噻唑蓝)摄入法检测Hep-2细胞增殖抑制率,流式细胞术检测细胞周期各时相细胞百分率变化及细胞凋亡率,电子显微镜观察亚细胞结构改变。结果:单纯热疗组、单纯TL组和TL联合热疗组Hep-2细胞增殖抑制率分别为21.2%、28.5%和54.5%,TL联合热疗组细胞增殖抑制率高于单纯热疗组和单纯TL组(P<0.01)。与对照组比较,单纯热疗组、单纯TL组和TL联合热疗组细胞周期进程发生明显改变,G1期细胞增多(P<0.01),S期细胞减少(P<0.01),而G2/M期细胞相对增多(P<0.01),细胞凋亡率升高(P<0.01);与单纯热疗组、单纯TL组比较,TL联合热疗组上述各项指标变化差异均具有显著性(P<0.01)。电子显微镜观察,单纯热疗组、单纯TL组和TL联合热疗组Hep-2细胞核染色质趋边凝集,线粒体肿胀,可见凋亡小体改变;上述变化TL联合加热疗组轻于单纯热疗组、单纯TL组;对照组未见上述改变。结论:TL联合热疗可提高Hep-2细胞的增殖抑制率、抑制Hep-2细胞G1期向S期转化进程,诱导Hep-2细胞凋亡。 Objective To explore the effect of triptolide(TL) combined with hyperthermia on proliferation and apoptosis of Hep-2 cells and its affect on cell cycle phases,and provide theoretical foundation for treatment of laryngocarcinoma.Methods Hep-2 cells at logarithmic growth phase were randomly divided into control,hyperthermia,TL,TL combined with hyperthermia groups.MTT method was adopted to investigate the proliferation inhibition rate of the Hep-2 cells.Cell cycle and apoptotic rate of Hep-2 cells were analyzed by flow cytometry,and then the changes of subcellular structures were observed by electron microscope.Results The proliferation inhibition rates in hyperthermia,TL,TL combined with hyperthermia groups were 21.2%,28.5% and 54.5%,respectively;the proliferation inhibition rate in TL combined with hyperthermia group was higher than those in hyperthermia and TL groups(P〈0.01).Compared with control group,the number of cells at G1 phase was increased(P〈0.01),the number of cells at Sphase was decreased(P〈0.01),the number of cells at G2/M phase was relatively increased(P〈0.01),the apoptotic rate was increased(P〈0.01) in hyperthermia,TL,TL combined with hyperthermia groups;compared with hyperthemia and TL groups,the indexes mentioned above in TL combined with hyperthermia group had significant differences(P〈0.01).The result of electron microscope indicated chromatin margination,mitochondrion swelling,apoptotic body formation in heperthermia,TL,TL combined with hyperthermia groups,no changes in control group.Conclusion TL combined with hyperthemia can inhibit the proliferation of Hep-2 cells,induce apoptosis of Hep-2 cells and arrest the Hep-2 cells at G1 phase.
