摘要
本研究从丝裂霉素 - C处理时间、消化时间及细胞浓度等方面比较了诸因素对小鼠胎儿成纤维细胞饲养层的影响。结果表明 ,1 0 μg/m L 丝裂霉素 - C处理以 1 .5~ 2 h为宜 ,0 .2 %胰酶 + 0 .0 4% EDTA消化时间以 2~ 3min( 37℃ )为宜 ,接种浓度以 3~ 3.5×1 0 5m L- 1为宜。在成纤维细胞的分离过程中 ,接种后 30 min换液 ,能达到纯化成纤维细胞的效果。
The influences of treating time with Mekemycin C,digesting time and concentration of feeding cells on MEF feeder layer were studied with murine embryos.The result showed as follows:①Treating with 10 μg/mL Mekemycin C,the time should be controlled within 1.5~2 h;②Digesting with 0.2% trypsin+0.04%EDTA,the time should be controlled within 2~3 min(37 ℃);③The concentration of feeding cells shhould be controlled withhin 3~3.5×10 5 mL -1 ;④During isolating MEF,the cells could be purified by changing Medium after 30 min feeding MEF on the plate.
