摘要
目的探讨昆明小鼠胚胎成纤维细胞分离与培养条件。方法取昆明小鼠胚胎消化分离制备细胞悬液,观察胰蛋白酶浓度、胎龄选择及接种浓度对成纤维细胞质量的影响。结果采用0.0625%胰蛋白酶分段消化孕13.5~14.5d胎鼠可制备足量、高活力的细胞悬液,以(30~40)×105/ml接种浓度接种时细胞生长状态较理想。结论昆明小鼠可分离成纤维细胞,用于滋养层制备。
Objective To study the method for isolation and culture of embryonic fibroblasts (MEFs) of Kunming mice.Methods Embryos of Kunming mice were digested for preparation of cell suspension,and the suitable concentration of trypsin was studied,and the influence of gestational age and inoculation concentration for fibroblasts were also studied.Results MEFs could be isolated from 13.5~15.5 d fetal mice by 0.0625% trypsin solution with digestion for several times. Under the same condition,fibroblasts at the density of (30~40)×10^5/ml grew much better.Conclusion MEFs can be isolated from Kunming mice and they may be applied for preparation of nutritional layers.
