氧化砷诱导人小细胞肺癌细胞凋亡的研究 被引量:2

摘要 目的 研究砷剂 (As2 O3 )对人小细胞肺癌细胞凋亡诱导作用以及干扰素 (IFNα 2b)对As2 O3 作用的影响。方法 采用DNA电泳、TUNEL原位末端标记和免疫细胞化学染色检测不同浓度As2 O3 作用不同时间对人小细胞肺癌细胞凋亡诱导作用及相关基因蛋白的表达。结果 在一定的浓度范围内 ,As2 O3 可诱导人小细胞肺癌细胞凋亡 ,并且随浓度的升高和作用时间的延长 ,凋亡细胞数增加 ,相关基因蛋白Bcl 2、PCNA表达降低。联合使用IFNα 2b可增强凋亡诱导作用。结论 As2 O3 可诱导人小细胞肺癌细胞凋亡 ,且与药物浓度和作用时间有关。IFNα Objective To study apoptosis of small cell lung cancer (SCLC) cell induced by As 2O 3 and enhancement effect of interferon (IFN) on As 2O 3. Methods DNA gel electrophoresis, terminal deoxynucleotidyl transferase nick end labeling (TUNEL) and immunocytochemistry were used to detect the apoptosis induced by As 2O 3 with different doses and time and the expression of Bcl 2 or PCNA. Results Within certain extent of concentration, As 2O 3 could induce apoptosis of SCLC cells. And apoptotic cells increased and expression of Bcl 2 and PCNA decreased with time prolonged or dose increased. The apoptosis inducing effect of As 2O 3 combined with IFN was much higher than that of As 2O 3 alone. Conclusion As 2O 3 may induce apoptosis of SCLC cells and the efficiency of apoptosis is related to dose and time. IFNα 2b might enhance the effect of apoptosis on SCLC induced by As 2O 3.
