利用逆转录巢式PCR技术检测 6 1例风疹高发人群的血液标本 ,经细胞培养技术从RVRNA阳性标本中分离风疹病毒株 ,应用细胞病变、血凝试验、电镜观察及PCR扩增等实验证实其为风疹病毒 ,命名为HRB株。同时还对该分离株的E1基因部分片段进行序列分析并与其它型别基因进行比较 ,结果显示 ,此分离株与中国疫苗株(BRDⅡ )及另外 2株香港分离株均不在同一分支 ,而是位于由 2 0世纪 6
Blood samples of 61 cases from rubella epidemic groups were detected by reverse transcription and nested PCR. A rubella virulent virus was isolated from RV RNA of the positive samples using cell culturing technique and named as HRB strain, which was then confirmed by cytopathic effect (CPE), hemagglutination test (HA test), electronic microscopy and PCR. At the same time, sequence analyses of part of E1 gene of the HRB and comparison with other type of gene were carried out. The result showed that the HRB was not on the same branch with Chinese vaccine strain (BRDII) and another two isolate strains from Hong Kong, but located on a sub branch constructed and isolated in 1960s.
