胃癌组织smad_4基因和转化生长因子β1及其受体的表达及意义 被引量:6

Expression of smad_4 mRNA,TGF-β1 and TGF-βR1 in gastric cancer tissue and its biological significance

摘要 目的研究胃癌组织中smad4mRNA、转化生长因子β1(TGFβ1)、转化生长因子β1受体(TGFβR1)的表达特征及其意义。方法常规石蜡包埋切片行smad4mRNA原位杂交染色及TGFβ1、TGFβR1免疫组织化学染色。结果smad4mRNA、TGFβ1、TGFβR1在胃腺癌、黏液腺癌和印戒细胞癌中阳性率无明显差异;组织学分级Ⅰ级、癌细胞未浸润至深肌层和淋巴结未转移的组织smad4mRNA、TGFβ1、TGFβR1阳性率明显高于组织学分级Ⅲ级、癌细胞浸润至深肌层或浆膜层和淋巴结转移组织,差异均有显著或非常显著性意义;smad4mRNA、TGFβ1、TGFβR1在胃癌中表达密切相关。结论smad4mRNA、TGFβ1和TGFβR13者表达与胃癌发生发展、生物学行为和预后可能有关,可作为重要的生物学标记物。 Objective To examine the expression of smad 4 mRNA,TGF-β1,TGF-βR1in gastric cancer tissue and its biological sig nificance.Methods Smad 4 mRNA was detected by in situ hybridiz ation technique,while TGF-β1and TGF-βR1were deteced by immunohistochemi cal method of avidin-biotin complex in formalin-fixed specimen s of 57patients.Results The expression of smad 4 mRNA,TGF-β1,TGF-βR1among adenocarcinoma,mucoadenocarcinoma and ring cell carcinoma wa s not significantly different.The positive rates of sma d 4 ,TGF-β1,TGF-βR1were significantly higher in the t issues of histological grade I,without infiltration into deep muscular layer and without lymphnode metastasis than t hat of histological gradeⅢ,with infiltration into deep muscul ar or serous layer and with lymphnode metastasis.There was a significant relationshi p of the expression among above three factors in gastric cancer tissues.Conclusions Smad 4 mRNA,TGF-β1and TGF-βR1may be associated with the carcinogenesis,biological behaviors and prognosis of gastric c ancer.These three factors may be use d as important biological markers in gastric cancer.
