摘要 目的:探讨Endostatin转基因双歧杆菌口服冻干粉剂(ETB-2)对裸鼠人胃癌模型的抑瘤效应,并探讨其抑瘤机制。方法:人胃癌细胞株MKN-45接种于BALB/L裸鼠建立动物模型;裸鼠随机分为6组,每组6只,采取灌胃给药。接种后根据肿瘤体积绘制肿瘤生长曲线;给药满4周后处死动物并计算抑瘤率。用免疫组化方法检测微血管密度(MVD)和增殖细胞核抗原(PCNA);用DNA原位末端标记(TUNEL)方法检测细胞凋亡。结果:对照组抑瘤率为0,MVD为5.8310.83,细胞凋亡率为1.30±o.1%;氟铁龙组抑瘤率为41.1%,MVD为5.70±0.67,细胞凋亡率为2.7%±0.2%。ETB-2高剂量次日给药组抑瘤率为47.5%,MVD为3.90±0.67,细胞凋亡率为13.9%±0.7%,2周给药组抑瘤率为43.2%,MVD为4.28±0.49,细胞凋亡率为13.9%±0.6%;低剂量次日给药组抑瘤率为36.0%,MVD为4.58±0.31,细胞凋亡率为7.7%±1.1%。结论:ETB-2可显著抑制胃癌生长,其机制可能是抑制肿瘤内微血管形成,使肿瘤细胞增殖下降,诱导肿瘤细胞凋亡,促进肿瘤细胞环死,其作用具有量效? Background/Aims: To investigate the influence of Endostatin transfected bifidobacterium oral powder preparation (ETB-2) on human gastric cancer and its mechanism of action. Methods: The hu- man gastric cancer cell line MKN-45 was implanted subcutaneously in BALB/L nude mice, then the mice were allocated into 6 groups at random, each group 6 mice. The ETB-2 was given by drenching. The tumor growth curve was got according to the tumor volume, and the rate of tumor inhibition was calculated after sacrificing the animals 4 weeks later. The microvessel density (MVD) and proliferating cell nuclear antigen (PCNA) index were measured immunohistochemically and the apoptotic rate was analyzed with terminal deoxynucleotidyl transferase mediated dUTP nick end labeling (TUNEL) tech- nique. Results: In the controls, the tumor inhibition rate was 0, the MVD was 5.83±0.83, the apopto- tic rate was 1.3%±0.1%, whereas in the fortulon group they were 41.1%, 5.70±0.67 and 2.7%±0.2% respectively. In the high dosage ETB-2 group where ETB-2 was given one day after implantation, the tumor inhibition rate was 47.5%, and MVD 3.90±0.67, the apoptotic rate 13.9%±0.7%; when ETB-2 was given two weeks later, the findings were 43.2%, 4.28±0.49 and 13.9%±0.6% respectively; in the low dosage group, ETB-2 was given one day after implantation, the findings were 36.0%, 4.58±0.31 and 7.7% ±1.1% respectively. Conclusions: ETB-2 can significantly inhibit the development of gastric can- cer, and its action is of dose-dependent, the mechanism of action may be due to its antiangiogenetic effect, inhibiting proliferation, also inducing apoptosis and necrosis.
