用mRNA差别显示方法分析黑麦盐胁迫下应答基因cDNA片段的表达特性 被引量:15

摘要 利用 m RNA差别显示方法分析经 2 5 0 mmol/ L Na Cl处理 3d的黑麦幼苗 ,14个差异 c DNA片段 ,包括 8个诱导表达片段 ,2个增强表达片段和 4个抑制表达片段被检测到。 Northern Blot分析证实 ,其中 0 .8kb、 0 .65 kb和 0 .35 kb c DNA片段有强烈阳性信号 ,命名为 SI80 0 、SI650 、SI350 (SaltInduced80 0 bp、 65 0 bp、 35 0 bp) ,证明与盐胁迫有关。以 SI80 0 为探针与分别经盐胁迫 3d、 7d、 14d的黑麦幼苗总体 RNA杂交 。 Fourteen differential expression cDNA fragments, including 8 inducti on expression fragments, 2 intensive expression fragments and 4 inhibition expre ssion fragments. have been detected in rye seedlings treated with 250mmol/L NaCl for 3d by using the techniques for differentially expressed mRNA display. North ern blot analysis indicated that 0.8 kb, 0.65 kb and 0.35 kb cDNA fr agments have ob viously hybrid signal, named SI 800 , SI 650 , SI 350 (Salt induced 800 bp, 650 bp and 350 bp). It thought to be related to salt stress. The expre ssion products were increased obviously at the differential salt stress time whe n total RNAs isolated from NaCl treated rye leaves for 3d, 7d and 14d respective ly, were hybridized to the SI 800 probe.
