摘要
目的 :研究肉苁蓉总苷 (Glycosides of Cistanche,GCs)对离体大鼠心脏缺血再灌注损伤的保护作用。方法:采用 L angendorff离体大鼠心脏灌流模型 ,在心脏低灌流期 (5 5 m in) ,给药组在灌流液中分别加入 GCs12 5、2 5 0 mg/ L,以普萘洛尔 (0 .2 μm ol/ L)为阳性对照组 ,以比色法测定心肌 Se- GSH- Px、SOD活性及丙二醛 (MDA)含量 ,电镜检查心肌超微结构的改变。 结果:GCs12 5、2 5 0 mg/ L组明显保护心肌 SOD、Se- GSH - Px活性 ,降低 MDA含量 ,增加再灌后冠脉流量 ,降低冠脉阻力 ,促进心肌收缩力的恢复 ,并明显减轻心肌超微结构损伤。结论:GCs可能是通过清除氧自由基 。
Objective: To investigate the effects of glycosides of cistanche (GCs) on the isolated reperfused rat's heart. Methods: The experiment examined the effects of GCs on myocardial ischemia reperfusion injury in isolated Langendorff perfused rat hearts. We used propanolol as positive contrast group. After stabilization, hearts were subjected to 55 min lower flow ischemia, immediately followed by 30 min reperfusion. GCs125 mg/L, 250 mg/L was perfused respectively in the period of ischemic arrest (0.3 ml/min). The activity of Se GSH Px, SOD and contents of MDA in myocardium were examined with colorimetry. The myocardial ultrastructure was observed with electron microscope. Results: GCs could improve cardiac functions, decreased the coronary vascular resistance, and increased the activity of SOD and Se GSH Px, reduced MDA content and protected the ultrastructure of myocardium from reperfusion injury. Conclusions: GCS have protective effects on myocardial ischemia/reperfusion injury probably by inhibiting lipid peroxidation.
