摘要
目的 :研究肉苁蓉总苷 (GCs)对异丙肾上腺素所致小鼠心肌损伤的保护作用。 方法 :采用异丙肾上腺素 (Isoprenaline,ISO)诱发小鼠急性心肌损伤模型。以比色法测定心肌 Se- GSH- Px、SOD活性及丙二醛 (MDA)含量。以生化自动分析仪测血清 CPK活性。电镜检查心肌超微结构的改变。 结果 :ISO(2 0 mg/kg皮下注射连续 2d)使心肌 Se- GSH - Px、SOD活性明显降低 ,MDA含量显著增加 ,并使心肌超微结构严重损伤 ,心肌释放 CPK增加。 GCs(12 5、2 5 0、5 0 0 mg/kg呈剂量依赖性地保护心肌 Se- GSH- Px、SOD活性 ,降低 MDA含量 ,减少 CPK释放 ,减轻 ISO引起的心肌超微结构损伤。 结论
Objective: To investigate the protective effects of glycosides of cistanche(GCs) against isoprenaline(ISO) induced myocardial damage of mice.Methods: The experiment studied the effects of GCs on myocardial injury model induced by ISO. The activity of Se GSH Px、SOD and contents of MDA in myocardium were examined with colorimetry. The activity of serum CPK was examined with automatic biochemistry analyzer. The myocardium ultrastructure was observed with electron microscope.Results:ISO (20 mg/kg,sc once a day for two days) caused widespread and severe myocardial cell damage on ultrastructural view, decreased myocardial Se GSH Px, SOD activities, increased myocardial MDA content and serum CPK concentration. Oral administration of GCs (125,250,500mg/kg) protected against these changes induced by ISO with a result of incressed myocardial Se GSH Px, SOD activity, decreased myocardial MDA content and serum CPK concentration. GCs(250,500 mg/kg) markedly reduced cardiotoxicity of ISO on the ultrastructural changes. Conclusions: GCs has protective effects aganist myocardial damage induced by ISO and for antioxidation.
