摘要
目的 :研究脑胶质瘤侵袭性生长过程中 ,C -myb的作用及其与蛋白激酶Ca 之间的相互关系。方法 :Wistar大鼠 35只 ,随机分为实验组 (30只 )和对照组 (5只 )。实验组每只大鼠于右侧大脑皮质运动区种植C6胶质瘤细胞 10 μl (3× 10 7个 / μl) ,对照组则注入 10 μl生理盐水 ,于术后 10天将大鼠全部处死取材 ,进行大体观察、HE染色及免疫组织化学检查 ,并将实验组侵袭性量化为 0~Ⅳ级 ,分别检查对照组和实验组肿瘤侵袭性中 ,各组C -myb、PKCa 的表达 ,并对其进行评分。结果 :1 实验组大鼠均可见肿瘤生长 ,其中 2 8只表现为侵袭 0~Ⅳ级 ;2 C -myb表达越强 ,脑胶质瘤侵袭的级别越高 ,呈正相关 (P <0 .0 1) ;3 肿瘤区C -myb与PKCa 表达在肿瘤各级中均为高度一致 ;4 在肿瘤 0~Ⅳ级侵袭中 ,瘤周区及边缘区的C -myb、PKCa 二者表达评分密切相关(P <0 . 0 5 ) ;5 对照组中 ,C -myb、PKCa 均为阴性表达。结论 :本组资料表明 ,C -myb是脑胶质瘤侵袭性的重要癌其因 。
Objective:To study the action of c-myb and the correlative relation among c-myb and PKCa in cerebral glioma invasive growth.Methods:Thirty five male Wistar rats were divided into experimental group(30)and control group(5)randomly.In experimental group,3×10 8cells/10ul c 6 glioma cells were implanted to the right cerebral cortex;in control group,10ul normal saline were injected in the same area as in experimental group.Ten days later,all rats were killed and given pathological examination.In experimental group,the glioma invasion was divided into I~IVgrades by gross observation and HE staining examination.With immunohistochemistry method,the expressing score of c-myb,pkc a were measured in the control group and in each invasive grade of the experimental group.Results:①all of them had tumor growth in the experimental group.Twenty eight rats have shown gliomainvasive I~IVgrade;②the more overexpressing the c-myb was,the higher was the glioma invasive grade.There was singnificant positive correlation between the c-myb expression scores and the glioma invasive grade( P <0 01);③in tumor area,every glioma invasive grades have high expressing unanimity between c-myb and PKC a;④When glioma invade as 0~IV grades,the expressing scores had close correlation among c-myb.PKC a in the area surrouding the tumor and at the verge of the area( P <0 05).⑤the c-myb. PKC a expressed negative in the control group.Conclusion:Our data had suggested that c-myb might be an inportant oncogene of tne glioma invasive growth.The invasive growth of cerebral glioma might be a main cause of recurrence of the glioma after surgery.
