摘要
目的 探讨原发性高血压 (EH)患者iNOS基因表达水平及其与ET、AngⅡ的关系。方法 5 5例EH患者 ,平均年龄 5 4.9± 9.2岁 ,男性 30例 ,女性 2 5例采用PCR技术检测淋巴细胞中的iNOS基因表达水平和放免法测定ET、AngⅡ血浆含量。结果 EH患者iNOS基因表达水平从Ⅰ、Ⅱ、Ⅲ期呈递增表达趋势 ;iNOS基因表达水平与年龄有关 ,年龄越大 ,增强趋势越明显 ,但无统计学意义。iNOS基因表达水平与ET、AngⅡ血浆含量呈正相关。结论 EH的病因可能与iNOS基因表达水平有关 ;EH患者iNOS基因表达水平与原发性高血压的病理生理改变关系密切 ,可以作为判定高血压程度的指标 ;血管活性物质AngⅡ、ET激活iNOS ,生成NO增多 。
Aim To investigate the relationship between expression of iNOS mRNA gene and plasma angiotensinⅡ(AngⅡ),endothelin(ET) in patients with essential hypertension. Methods 55 cases patients with essential hypertension were divided into 3 groups (stageⅠ17cases、stageⅡ 24cases 、stage Ⅲ14 cases). 24 normal individuals matched in age and heart rate and body mass index served as a normal controls(NC). The expression of iNOS mRNA gene in lymphocytes was measured by polymerase chain reaction(PCR) and AngⅡ,ET were detected by radioimmunoassay. Results (1)The level of mRNA of iNOS in essential hypertension were significantly higher than that NC group, The increase in mRNA of iNOS were more obvious in stage Ⅱ,Ⅲ EH than that in stageⅠ( P< 0.01). (2)Plasma AngⅡ,ET in EH were higher than those in NC group( P< 0.05 and P< 0.01); however, the difference have not statistical significance in stage Ⅰ、Ⅱ、Ⅲ EH ( P >0.05). Conclusion These data strongly indicate that higher expression of iNOS mRNA in lymphocytes in patients with essential hypertension may play regulatory role for hypertension.
