摘要
应用反转录-聚合酶链式反应(reversetranscription-polymerasechainreaction,RT-PCR)方法,测定出一氧化氮合成酶(nitricoxidesynthetase,NOS)mRNA不仅在心血管系统细胞中广泛存在,并且发现内皮素可以促进血管平滑肌细胞中NOSmRNA的表达(+34.8%),增高血管平滑肌细胞中NOS酶活性(+74%)。结果提示,由内皮素引起的血管平滑肌细胞中NOS基因表达和NOS活性的增加,可能是高血压中的一个重要的负反馈因素。
With reverse transcripti0n-polymerase chain reaction (RT-PCR),it has been found that NOS mRN;\ is widely distributed in cells of car<liovascular system such as t'ardiomyocytes, endothelia1 ce]ls, vascular smooth musclc cel1s (VSMC ). When ET (1O-' mol/L ) was added to cu1tured VSMC, it significant1y increased the NOS mRNA leve1 in VSMC (+ 34. 8% ). By measuring the enzyme activity of NOS in cultured VSMC,it was f0und that when VSMC was treated with ET (2 X 10wv mol/L) l NOS activati0n was significantly increased (79% ) compared to the contr0l. These data show that the increased expression of NOS gene inVSMC induced by ET may be an important regulat0ry factor for hypertension.
