成束蛋白在乳癌的表达及其与雌激素受体的相互关系和意义 被引量:3

摘要 目的 探讨与细胞运动相关的成束蛋白在原发性乳癌的表达 ,及其与雌激素受体(ER)阴性癌细胞的关系。方法 选取乳癌标本共 84例 ,用免疫组织化学法检测成束蛋白的表达情况 ,并与ER和增殖细胞核抗原 (PCNA)染色状况相比较。结果 成束蛋白主要在癌细胞胞浆内弥漫性着色 ,在 5 3例ER阳性病例中仅 3例为成束蛋白表达阳性 (5 .7% ) ,相反 ,在31例ER阴性肿瘤中有 2 1例表达阳性 (6 7.7% ) ,二者差异显著 (P <0 .0 0 1) ;但成束蛋白表达与绝经状态、肿瘤大小、组织学分级和淋巴结转移状况等无显著相关 (P >0 .0 5 ) ;然而 ,阳性成束蛋白表达与PCNA表达呈显著的正相关 (P <0 .0 0 1)。结论 人成束蛋白的过度表达可导致细胞间粘着力的降低和细胞运动活性的增加 ,可能是由于肌动蛋白动力学中的分子调节对癌细胞的恶性表型具有一定的影响 ;而肌动蛋白的功能作用受肌动蛋白相关蛋白 ,如成束蛋白的活性控制。且成束蛋白大多存在于那些高表达PCNA的癌细胞 ,也可能与高增殖活性的癌细胞有关 ,这可能是阴性ER乳癌相对于阳性ER组有着更差的预后的原因之一 ,但具体作用机制和临床意义还有待于进一步研究。 Objective To study the expression of fascin, an actin bundling protein associated with cell motility, and the relationship with ER negative breast carcinoma.Methods Comparasion of the expression of fascin, estrogen receptor (ER) and proliferative cell nuclear antigen (PCNA) determined by immunohistological method was made on 84 specimens of primary breast carcinomas. Results The diffuse staining of fascin in cancerous cell cytoplasm was noted. Only 3 of 53 breast carcinomas with ER positive showed fascin positive expression (5.67%); in contrast, 21 of 31 carcinomas with ER-negative showed positive expression(67.7 %) (P< 0.001). Whereas fascin expression showed no significant correlation with menopause status, tumor size, histologic grading or lymph node metastasis. However, fascin expression was highly related to the expression of PCNA in cancer cells (P< 0.001 ). Conclusions Over expression of fascin could induce decreased intercellular adhesion and increased cell motility, which might be attributed to the molecular modulators of actin dynamics anticipated to the malignant phenotype of cancer cells. Actin function is influenced by action-bundling associated protein such as fascin, in additioin, fascin is mainly presented in highly PCNA-expressed cancer cells, which suggests that such cancer cells are related to those with high proliferative activity, and may be one of the reasons that the patients with ER-negative breast cancer has worse prognosis than those with ER-positive cancer. But the real mechnism and clinical significance of fascin need futher studies.
