摘要
利用孕酮 (Progesterone ,P4 )单克隆抗体 ,建立P4 自包被固相放免分析法 (Self -coatingRadioim munoassay,Sc -RIA)。其灵敏度为 0 .0 6nmol/L ,回收率达 95 .73 %批内CV6.8% ,,批间CV9.3 %。对 5头正常健康未孕奶牛的一个发情周期的奶样进行检测 ,检测范围在 0 .0 9～ 2 1 .3 7nmol/L ;同时对 2 0头配种奶牛进行早期妊娠诊断 ,妊娠诊断准确率达 89.4 7% ,未孕诊断准确率达 1 0 0 %。
The self-coation solid-phase radioimmunoassay(S c-RIA)of progesterone is developed with progesterone monoclonal antibody.The sen sitivity is 0.06nmol/L.The average recovery is up to 95.73%.The intra-and inter -assay coefficient of variation are 6.8% and 9.3%.The skim milk progesterone le vels of 5 control milk cows range from 0.09 to 21.37 nmol/L.The result of preli minary early pregnancy diagnosis shows that accuracy of the pregnant and the non -pregnant are 89.47% and 100%.
