摘要
本试验选用单克隆抗体采用非平衡法建立起孕酮的酶免疫分析法.整个测试过程在1小时内完成,结合率与孕酮浓度的自然对数的典型的回归方程式为:E/E_0=38.84LnP—21.53(r=-0.9763),灵敏度为0.05n mol/L.标准曲线的可测范围为0~3.2n mol/L.标准曲线7个剂量点的平均批间变异系数为4.75%,同一样品批内和批间变异系数分别为6.8%和10.25%,回收率为101.10±4.35%.孕酮单克隆抗体与7种甾体激素的交叉反应率均小于0.5%.该法测定脱脂乳样品所得的结果与用常规血清抗体建立的酶免疫分析法所得结果密切相关(r=0.9551).
An enzymeimmunoassay (EIA) for progesterone is described by using monoclonal antibody with non-equilibrium method. A lower limit of sensitivity of 0. 05n mol/L was obtained with the response being linear (Logit/Ln transformation) through 3. 2nmol/L. The typical regression equation of dose-response curve is: E/ Eo = 38. 84 LnP-21. 53 (r= -0. 9763). Results obtained by this EIA and ordinary EIA in defatted milk samples gave excellent agreement (r=0. 9551). This assay system allows accurate determination of progesterone with good specificity, precision, accuracy, and rapidity requiring less than one hour to perform, and is suitable for the routine tests in any laboratory, even on the farm.
