亚低温对肝缺血预处理保护作用的增强效应

摘要 目的 探讨肝缺血再灌注损伤中亚低温 (MH )对缺血预处理 (IP)保护作用的增强效应机理。方法 观察非缺血对照组、缺血再灌注组、缺血预处理组和亚低温缺血预处理组 4组犬肝上下腔静脉血谷丙转氨酶 (ALT)、谷草转氨酶 (AST)、乳酸脱氢酶 (LDH )以及丙二醛 (MDA)和超氧化物歧化酶 (SOD)、过氧化氢酶 (CAT)、谷胱甘肽过氧化酶(GSH PX)活性及总抗氧化 (TAX )能力变化。结果 肝缺血再灌注后ALT、AST、LDH和MDA含量明显上升 (P<0 .0 1) ,SOD、CAT、GSH PX活性及TAX能力明显下降 (P<0 .0 1) ;缺血预处理组与缺血再灌注组比较及亚低温缺血预处理组与缺血预处理组比较 ,ALT、AST、LDH和MDA含量均明显下降 (P<0 .0 1,P<0 .0 5 ) ,SOD、CAT、GSH PX活性及TAX能力明显上升 (P<0 .0 1,P<0 .0 5 )。结论 缺血预处理能增强肝组织自身抗氧化能力 ,减轻肝缺血再灌注后氧自由基对肝脏的损伤 ;亚低温对缺血预处理的这种保护作用具有增强效应。 Objective To study the mechanism s of enhancing effect of mild hypothermia (MH) to ischemic preconditioning (IP) on h epatic ischemia-reperfusion (I-R) injury. Methods To observe the content of the marker e nzymes of liver damage (ALT,AST,LDH) and malondialdehyde (MDA), and activities of superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GSH-PX), total antioxidase (TAX) in inferior vena cava blood above liver in nonischemic control group (n=6), I-R group (n=6), IP group (n=6) and mild hypo thermic ischemic preconditioning (MHIP) group (n=6). Results After I-R the content of ALT,AST, LDH and MDA were significantly elevated (P<0.01), SOD,CAT ,GSH-PX,ACT activities were declined obviously (P<0.01). The content of ALT,AST,LDH and MDA were significantly lower in IP grou p than those in I-R group, and in MHIP group than those in IP group (P<0.01,P<0.05), and the content of SOD, CAT,GSH-PX, ACT activities were significantly higher in IP group than those in I-R group, and in MHIP group than those in IP group (P< 0.01,P<0.05). Conclusion Ischemic preconditioning may enhanc e the oxidation-resistance of liver, and reduce the oxygen free radical injur y to liver after ischemia-reperfusion. Mild hypothermia may enhance the protect ive effect of IP on hepatic ischemia-reperfusion injury.
