摘要
目的:探讨重组人促红细胞生成素(Recombinant human eryfhropoietin,rHuEPO)对脂多糖(Lipopolysaccharide,LPS)诱导的人脐静脉内皮细胞(Human umbilical vein endotheilial cells,HUVECs)凋亡的影响。方法:实验分为四组;第一组仅用LPS处理;第二组预先用rHuEPO孵育后再用LPS处理;第三组只用rHuEPO处理;对照组用无血清的M199培养。用流式细胞仪DNA分析检测细胞凋亡。结果:与对照组相比,LPS处理后的细胞DNA亚二倍体含量显著增加(P<0.05),预先用rHuEPO处理后则可显著降低LPS的这种作用(P<0.05)。结论:rHuEPO抑制了LPS诱导的内皮细胞凋亡,这种保护作用可能是rHuEPO诱导内皮细胞生长和血管生成的一个重要因素。
Objective: To investigate the inhibition of recombinant human erythopoietin (rHuEPO) on lipopolysaccharide (LPS) induced apoptosis in human umbilical vein endothelial cells (HUVECs). Methods: The experiments were divided into Four groups. The first group was incubated with only LPS. The second group was preincubated with rHuEPO follow by LPS. The third group was treated with only rHuEPO. Control cells were cultured in serum-free medium. Apoptosis was determined by flow cytometric DNA analysis. Results: Compared with control group, the hypodiploid DNA was increased sigificantly on HUVECs treated with only LPS (P<0.05), preconditioning with rHuEPO decreased markly the content of hypodiploid DNA (P<0.05). Conclusions: rHuEPO inhibit LPS-induced apoptosis in endothelial cells, this effect may play an important role in rHuEPO-induced endothelial cell growth and angiogenesis.
