摘要
用混合酸苷法将脱落酸(ABA)与卵清蛋白(OA)连接,形成的ABA-OA复合物用于包板,利用兔抗ABA抗体与样品中或标准品中的ABA及板上的ABA反应。建立了ABA间接酶联免疫检测法(ELISA)。本法的检测范围为0.122~125pmol,检测灵敏度为0.02pmol,并具有良好的健全性。植物材料的激素提取液经初步纯化即可用于测定。用本法对正常供水及缺水1周的鸭趾草叶片中的ABA含量进行了检测,干旱处理的(26pmol/gFW)为正常供水(9.3pmol/gFW)的3倍,而且干旱处理的叶片下表皮的ABA含量(37.5pmol/gFw)比上表皮的(17.7pmol/gFW)高2倍。
ABA-OA conjugates were synthesized by linking ABA to ovalbumin (OA) with mixed anhydride. An indirect method of enzyme linked immunosorbent assay (ELISA) was established by utilizing ABA-OA conjugates for coating wells, on which rabbit anti-ABA sera reacted. Sensitivity and detection range of the assay were 0.02 pmol and 0.122~125 pmol, respectively. Plant extract from 50~500mg samples was easy to be prepared. ABA level of lcaves of Commelina communis L. was quantitatively determined with ABA indirect ELISA. ABA contcnt of the normally watered leaves was 9.3 pmol/g FW, lower than that of dry treated leaves (26 pmol/g FW). ABA level of lower cpidcrmis of dry treated leaves (37.5 pmol/g FW) was two times as high as that of upper epidermis (17.7 pmol/g FW).
