摘要
目的 :探讨不同浓度的干扰素 (IFN α)对人口腔表皮癌细胞株KB及多药耐药细胞株KBv2 0 0的影响。方法 :以MTT法测定梯度浓度的干扰素及长春新碱 (VCR)的细胞毒性变化。以流式细胞仪测定IFN α作用后肿瘤细胞对罗丹明蓄积作用的变化及P 膜糖蛋白 (Pgp)的变化。用RT PCR法检测多药耐药基因 (mdr 1)mRNA。 结果 :高浓度IFN α(10 0 0 0IU/ml)能明显抑制KB/KBv2 0 0细胞的增殖作用。低浓度IFN α(10 0 0IU/ml)能增强VCR对耐药细胞KBv2 0 0的杀伤作用 ,增加KBv2 0 0细胞对罗丹明的蓄积 ,下调Pgp和mdr 1mRNA表达 ,对敏感细胞KB无作用。 结论 :IFN α对KBv2 0 0细胞的多药耐药性有明显的逆转作用 ,且在一定浓度范围内有剂量依赖性 ,其机制是通过下调Pgp和mdr 1mRNA表达逆转多药耐药性。
Objective: To explore the effects of various concentrations IFN-α on human oral epidermic cancer cell line KB and its multidrug resistant counterpart KBv200.Methods: The toxicity of IFN-α and/or VCR was assayed by MTT method; intracellular rhodamin123 concentration and p-glycoprotein expression were measured by flow cytometry; mdr-1 mRNA was assayed by RT-PCR. Results: High-dose IFN-α(10 000 IU/ml) had significant antiproliferative effects on KB/KBv200 cell lines. Low-dose IFN-α did not have any significant effect on growth of KB/KBv200 cells, but did increase the cytotoxic effect of VCR and the accumulation of Rhodamine123 in KBv200 cell line and had no effect on parent VCR-sensitive KB cell line. IFN-α down-regulated expression of P-gp and mdr-1mRNA . Conclusions: IFN-α has significant effect on the reversal of multidrug resistance of KBv200 cell line via a mechanism of P-gp and mdr1 mRNA down-regulation.
