IL-6、IL-1β、TNFα对人胎大脑神经细胞体外生长的影响 被引量:3

摘要 目的 :研究IL 6、IL 1β、TNFα在人胎大脑神经细胞体外发育过程中的作用 ,为临床防治提供实验依据。 方法 :应用 10~ 13周龄人胎大脑神经细胞无血清原代培养模型 ,观察活细胞形态 ,MTT法测定细胞存活率。结果 :IL 6促进细胞贴壁、突起生长与伸展 ,IL 1β、TNFα对形态学影响不大。IL 6、IL 1β单独作用时 ,细胞存活率呈一过性下降 ,后又出现升高 ,但IL 6升高出现较早 ,作用较强 ,作用强度与实验浓度无关 ;TNFα( 10~ 10 0 0U/ml)作用下细胞存活率持续低于对照组 ,仅 10U/ml组体外 16d( 16DIV)时高于对照组 10 8 2 6% ;IL 1β与TNFα联合作用时仅于 16DIV时为对照组 10 9 90 % ( P <0 0 1) ,呈轻度协同效应。结论 :人胎大脑神经细胞体外发育过程中 ,IL 6、IL 1β具有神经营养作用 ,促进细胞存活 ,IL 6还可促进细胞生长、分化 ;TNFα体外主要为神经毒性作用 ;IL 1β与TNFα联合作用于培养后期可显轻度协同效应。作用机理可能与局部自分泌与旁分泌有关。 Objective:To study the effects of Il 6?IL 1β?TNFα on the growth and development of cultured human fetal cerebral neurons.Methods:Established the model of primary culture of human fetal cerbral neurons in SFFD,by morphological observation and MTT assay.Results:IL 6 and IL 1β both promote the survival of neurons and IL 6 also promote the outgrowth of neuritis and differentiation of neurons in culture.TNF α continuously inhibit the survival of neurons.Conclusion:IL 6?IL 1β?TNFα can effect the development of cultured neurons.IL 6 and IL 1β both may effect as neurotrophic factor in the survival?growth and development by indirect mechanism.TNFα directly show neurotoxin effect in culture.
