目的··:探讨5 -羟色胺选择性重摄取抑制剂 (SSRI)氟西汀和帕罗西汀对甲基苯丙胺 (MA)诱导的条件性位置偏爱效应(CPP)的影响。方法·· :采用倾向性实验程序。♂Wistar大鼠腹腔注射MA(0.5mg·kg-1)并训练8d ,d9测定大鼠对伴药盒的偏爱效应及测试前30min注射不同剂量 (2.5 -10.0mg·kg-1)氟西汀和帕罗西汀对该效应的影响。结果··:0.5mg·kg-1 的MA可以诱导大鼠对伴药盒产生显著的条件性位置偏爱 ;测试前注射氟西汀和帕罗西汀均可剂量依赖性地降低MA诱导的条件性位置偏爱效应的表达。结论··:氟西汀和帕罗西汀对MA的强化效应有抑制作用 。
Objective: To study the effect of serotonin selective reuptake inhibitors (SSRI) fluoxetine and paroxetine on the methamphetamine (MA)-induced conditioned place preference (CPP) in rats. Method: A biased procedure was adopted in the present study. Male Wistar rats were given MA (0.5 mg·kg-1,ip)and trained for 8 days. The preference for the drug-paired box of the rats and the effect of different doses of fluoxetine or paroxetine (ip) pretreatment before test on the CPP response were determined on the 9th day. Result: MA 0.5 mg·kg-1 appeared to be enough for rats to obtain a significant place preference for the drug-paired box. Pretreatment with 2.5,5.0 and 10.0 mg·kg-1 of fluoxetine and paroxetine before test inhibited the expression of this response in a dose-dependent manner. Conclusion: Fluoxetine and paroxetine inhibited the reinforcement effect of MA, indicating that this kind of drug might be potentially useful in the treatment of methamphetamine psychological dependence.
