摘要
目的 :建立重组人成纤维细胞生长因子的等电聚焦电泳法。方法 :利用载体两性电解质和聚丙烯酰胺 ,采用毛细管灌胶法自制平板胶 ,并从凝胶浓度、电极溶液等方面优化重组人成纤维细胞生长因子的等电聚焦电泳条件。结果 :pH梯度曲线线性回归方程为Y =- 1 2 5 93X + 10 80 8,r =0 993 9。重组人成纤维细胞生长因子的等电点为 9 4(n =5 ,RSD =1 0 % )。结论 :该法简便 ,实用 ,重复性好 ,为碱性多肽或蛋白的等电点测定提供了实验依据。
Objective:To establish the isoelectrofocusing (IEF) method of rhbFGF.Methods:Horizontal gel is prepared by using Amphline and acrylamide and the condition of electrophoresis is optimized.Results:Isoelectric point of rhbFGF is 9 4( n=5, RSD=1 0%) and the linear equation of standard proteins is Y=-1 2593X+10 808,r=0 993 9. Conclusion:This method is simple and has good reproduction,it provides foundation of experiments for IEF of basic polypeptides and proteins.
