兔脑出血灶周围脑组织细胞凋亡及bcl-2和bax的检测 被引量:4

摘要 目的 :探讨兔脑出血灶周围脑组织中bcl 2、bax蛋白表达和细胞凋亡的关系。方法 :家兔 40只 ,分为A、B、C、D 4组。B、C、D 3组采用兔自体血二次注射法制作脑出血模型 ,分别于模型制作完成后 12h ,2 4h ,72h断头处死 ,运用TUNEL法、免疫组化SP技术 ,检测出血灶周围脑组织中细胞凋亡及bcl 2、bax蛋白表达。结果 :出血灶周围脑组织中细胞凋亡、bcl 2和bax蛋白表达在各组间差异有显著性 (P <0 .0 5 )。bcl 2蛋白表达与凋亡阳性细胞数呈负相关 (r=- 0 .781,P <0 .0 5 ) ,bax蛋白表达与其呈正相关 (r=0 .72 3,P <0 .0 5 )。结论 :出血灶周围脑组织中存在细胞凋亡 ,细胞凋亡的变化在一定程度上反映了周围脑组织的损伤程度。bcl 2和bax蛋白对凋亡具有调控作用。 Aim: To investigate the relations between the expressions of bcl 2, bax protein and apoptosis in perihematomal brain regions of rabbits. Methods: A total of 40 rabbits were equally divided into 4 groups,group B,C and D were experiment groups,and group A,normal control group.Intracerebral hemorrhage model was made by double injection of self body blood.Rabbits in group B,C and D were executed after 12 h,24 h,and 72 h,respectively.TUNEL method was used to detect apoptosis,and SP method to detect expressions of bcl 2 and bax protein in the perihematomal brain tissues.Results: The apoptosis and expressions of bcl 2 and bax protein among experiment groups were significantly different ( P< 0.05).The expression of bcl 2 protein was negatively correlated with the number of apoptotic cells ( r=-0.781, P< 0.05), and the expression of bax protein was positively correlated with apoptotic cells ( r=0.723, P <0.05).Conclusions: It is indicated that apoptosis may present and reflect brain damage after intracerebral hemorrhage. Gene bcl 2 and bax may play an important role during the period of apoptosis.
