摘要
目的研究表达自杀基因的减毒鼠伤寒沙门氏菌对肿瘤的抑制作用。方法把表达自杀基因的质粒pcCD导入减毒的鼠伤寒沙门氏菌,以荷瘤鼠为模型研究沙门氏菌/自杀基因与5FC系统对肿瘤的抑制作用。结果该系统能抑制肿瘤生长,延长荷瘤鼠的存活期。结论鼠伤寒沙门氏菌可以作为基因治疗的候选载体。
Aim To research attenuated Salmonella typhimurium as suicide gene delivery vector in tumor retard ation.Methods The plasmid pc-CD which express suicide gene is transferred into attenuated Salmonella ty-phimurium by electric transperforation.We use the tumor-bearing nude mice as tumor animal model to researc h the tumor-retarded effect of the system including the auxotrophic mu tants containing the suicide gene CD and the prodrug 5-FC.Results The system can suppress tumor growth and prolong the survival of the tumor-bearing nude.Conclusion Salmonella typhimurium can be the candidate gene therapy vector.
