摘要
目的 研究白细胞介素 - 6 (IL - 6 )对受分次照射的荷瘤小鼠肿瘤细胞凋亡的影响 ,探讨IL - 6辐射防护与肿瘤增敏效应的机制。方法 2组荷瘤小鼠受 8Gy/ 1Gy/ 8d照射后 ,IL - 6组尾静脉注射IL - 6 40 0 μg/ (kg·d) ,对照组注射同剂量白蛋白。将小鼠活杀 ,无菌条件下取肿瘤组织 ,立即制备单细胞悬液后流式细胞术PI染色法检测肿瘤细胞凋亡。结果 IL - 6组与对照组比较 ,肿瘤细胞凋亡指数无明显差异 (P >0 .0 5 )。结论 IL - 6无促进受分次照射的荷瘤小鼠肿瘤细胞凋亡的作用 ,IL - 6通过激发荷瘤宿主对肿瘤细胞的免疫反应 。
Objective To study the effect of IL-6 on the apoptosis of tumor cells in irradiated tumor-bearing mice and to discuss the mechanism of radioprotective effect and radiosensitizing effects of IL-6. Methods Two gropups of tumor-bearing mice were subjected to fractional irradiation (8Gy/1Gy/8d). IL-6 400 μg/(kg.d) iv was administer to the mice in the test group, with similar dose of albumin given to the control mice. By the end of the experiment, the mice were killed to get the carcer tissue immediately prepared into single-cell suspensio and stained with PI to assess the apoptosis of tumor cells by using FCM. Results The apoptosis index (AI) showed no significant difference between IL-6 test group and control group(P> 0.05 ). Conclusion IL-6 does not stimulate the apoptosis of carcinoma cells and its radioprotective and radiosensitization roles shoud be played by stimulating the immuneresponse of tumor-bearing mice to carcinoma.
