摘要
目的 探讨钩端螺旋体外膜蛋白基因在钩体遗传分类中的作用。方法 对我国的 2株问号钩体赖型 5 6 6 0 1株和爪哇型 5 6 6 0 2株的外膜蛋白基因Ompl1进行PCR扩增 ,得到外膜蛋白基因并进行了全基因序列分析。测序的 2株钩体与流感伤寒型RH5 2株进行了核苷酸序列比较和推导的氨基酸序列比较。结果 赖型 5 6 6 0 1株和爪哇型 5 6 6 0 2株钩体Ompl1基因核苷酸序列与流感伤寒型RH5 2株的同源性分别为 89%和 80 % ,推导的外膜蛋白氨基酸序列的同源性分别为 92 %和88%。由此可以看出 5 6 6 0 1株、5 6 6 0 2株与RH5 2株分别属于不同的血清群 ,这与Yasuda的遗传分类相一致。结论 我国的赖型 5 6 6 0 1株和爪哇型 5 6 6 0 2株及流感伤寒型RH5 2株Ompl1基因的核苷酸序列存在一定差别 。
Aim The nucleotide sequences of Ompl1 geng of L.interrogans serovar lai and Javanica was analyzed.Methods The Ompl1 genes of two strains was obtained by PCR and the sequences analysis was conducted.Their nucleotide sequences and deduced amino acid sequences was compared with serovar grippotyphosa.Results The sequences of the Ompl1 gene of 56601 and 56602 strains showed homologous of 89% and 80% with RH52 strain and their deduced Ompl1 protein sequences had homologous of 92% and 88%.Conclusion The sequences of Ompl1 gene of different serogroups are distinct form each other and further research should be done on Ompl1 gene.
