摘要
目的 探讨胰岛素生长因子 1(IGF 1)在实验性脑出血大鼠急性期血浆和脑组织中的含量变化 ,阐明在脑出血过程的作用 .方法 SD大鼠 80只 ,随机分为对照组、小量脑出血组、中量脑出血组和大量脑出血组 ,每组 2 0只 ,每组按出血后 1,8,2 4,48和 72h再分成 5小组 ,每小组 4只 .采用Nath法建立大鼠脑出血模型 ,用放射免疫法测定血浆和脑组织中IGF 1的含量 .结果 与对照组相比 ,大 ,中 ,小量脑出血组在脑出血后 1h血浆中的IGF 1量开始减少 ,但无显著差异 ,8,2 4和 48h下降明显 (P <0 .0 1) ,以 8h下降最明显 ,72h已恢复至 1h水平 ,与对照组相比 ,脑出血后 1,8,2 4和 48h血肿周围脑组织中IGF 1的含量明显增高 (P <0 .0 1) ,8h为峰值 ,以后逐渐下降 ,72h接近对照组 .大 ,中 ,小量脑出血组在8,2 4和 48h时象相比有非常明显差异 (P <0 .0 1) .结论 IGF 1可能参与了脑出血的病理生理过程 。
AIM To observe insulin like growth factor 1 (IGF 1) concentration change in plasma perihemotoma of rats with acute cerebral hemorrhage in order to explore the effect of IGF 1 on acute cerebral hemorrhage. METHODS Eighty SD rats weighted 250~300 g were randomly assigned to four groups: control group, and little, medium, largequantity hemorrhage groups with each group divided into 1, 8, 24, 48 and 72 h post hemorrhage subgroups. The model of cerebral hemorrhage was copied according to Nath's and IGF 1 contents in rat plasma and perihemotoma were measured with radioimmunoassay. RESULTS Compared to control group, plasma IGF 1 content began to decrease after acute cerebral hemorrhage, with no significant difference at 1 and 72 h, significant difference ( P <0.01) at 8, 24 and 48 h post cerebral hemorrhage, especially at 8 h; and with no significant difference at the same time points during three experiment groups. Compared to control group, IGF 1 contents in perihemotoma appeared statistically significantly higher during 48 h post acute hemorrhage ( P <0.01), at the peak at 8 h, but 72 h post hemorrhage not significantly higher. Among three experiment groups, IGF 1 contents in large quantity hemorrhage group appeared significantly higher than that in little and medium hemorrhage ones from 8 to 48 h post hemorrhage ( P <0.01). CONCLUSION IGF 1 might be involved in the pathogenesis of acute cerebral hemorrhage.
