摘要
目的:研究肝素和氢化可的松以及二者联合应用对人脐静脉内皮细胞(ECV304细胞)和成纤维细胞(WI-38细胞)的生长抑制作用和细胞凋亡的影响,并比较普通肝素与低分子肝素作用的差异,探讨肝素和氢化可的松联合应用抑制血管生成的可能性。方法:将细胞在含有药物的培养液中孵育一定时间,观察其形态,用MTT法测定细胞的增殖抑制率,电镜下观察细胞凋亡情况和ELISA法定量检测细胞凋亡程度。结果:肝素和氢化可的松单独均可抑制ECV304细胞增殖,作用呈剂量依赖性;肝素对WI-38细胞亦有增殖抑制作用。肝素和氢化可的松联合应用具有协同作用。低分子肝素抑制作用强于普通肝素。经电镜观察和ELISA法检测,肝素和(或)氢化可的松没有明显诱导细胞凋亡作用。结论:肝素可抑制血管内皮细胞增殖但不具有特异性,且和氢化可的松具有协同作用。以上作用不是通过诱导细胞凋亡实现的。
Objective To study the proliferation-inhibiting and apoptosis-inducing effects of heparin and/or hydrocortisone to both human umbilical vein endothelial cell ECV304 and fibroblast WI-38.to compare the different effects between unfractionated and low-molecular-weight heparin.and to investigate the possibility of combined application of heparin and hydrocortisone in angiogenesis inhibition. Methods The different cells were cultured for a period of time in different kinds of culture fluid including different drugs. We observed the morphology of the cells under optic and electronic microscopes.and investigated the proliferation-inhibiting ratio and apoptosis degree by using MTT-cell proliferation detection and ELISA-cell death detection methods. Results Heparin or hydrocortisone inhibited ECV304 cell proliferation in a dose-dependent manner separately.so did heparin to WI-38 cells. Combination of heparin and hydrocortisone showed synergetic effect. And low-molecular-weight heparin showed powerful inhibition effects than unfrationated heparin. No obvious apoptosis-inducing effects of heparin and/or hydrocortisone was found. Conclusion Heparin inhibits the proliferation of vascular endothelial cells unspecifically and has a synergetic effect with hydrocortisone. The inhibition effect does not depend on apoptosis-inducing effect.
