摘要
目的观察高功率聚焦超声(HIFU)联合肝素可的松治疗裸鼠皮下LTNM人肝癌疗效。方法HIFU照射采用频率为1.1MHz,功率750W/cm2,连续超声波照射时间20秒。HIFU照射后第1天起,肝素以300U/ml饮水口服,可的松以初始剂量250mg/kg作皮下注射。48只裸鼠肝癌模型随机分为四组:Ⅰ组(n=12)为对照组;Ⅱ组(n=12)为肝素可的松治疗组;Ⅲ组(n=12)为HIFU照射组;Ⅳ组(n=12)为HIFU+肝素可的松治疗组。结果Ⅱ、Ⅲ、Ⅳ组均显著抑制肿瘤生长,治疗后第28天,抑瘤率分别为86%、90%和99%。Ⅳ组与Ⅱ、Ⅲ组比较,差异均有显著性(P<0.05)。Ⅳ组在治疗后35天肿瘤均消退,组织学检查示仅存疤痕组织。结论HIFU能有效破坏肝癌组织,HIFU辅以血管生成抑制剂治疗肝癌是一种有效彻底的综合治疗模式。
Objective To assess the efficacy of high intensity focused ultrasound (HIFU) combined with heparin and cortisone acetate in treating primary liver cnacer LTNM implanted subcutaneously in the nude mice.Method HIFU treatment was administered with a focused 1.1MHz transducer at a peak intensity of 750W/cm 2 for continuous exposure duration of 20s, and one day later, heparin (300U/ml) was given orally in the drinking water and cortisone acetate of 250mg/kg as well. The initial dose was injected subcutaneously per mouse. 48 mice with tumor were randomly divided into four groups. Group Ⅰ( n =12) as controls; group Ⅱ( n =12) received heparin and contisone therapy; group Ⅲ( n =12) received HIFU; group Ⅳ( n =12) received both HIFU and heparin plus cortisone. Results Significant inhibition of the tumor growth was seen in group Ⅱ,Ⅲ and Ⅳ as compared with group Ⅰ( P <0.05), with tumor growth inhibitive rate at the 28th day being 86%,90% and 99%, respectively. All mice in group Ⅳ were cured with no evidence of tumor at 35 days and no tumor cell left histologically. Conclusion The data suggest the HIFU can destroy liver cancer effectively and HIFU combined with antiangiogenic agent heparin and cortisone may be a mode for curative comprehensive treatment.
