摘要
目的 了解重组人肝再生增强因子对实验性肝纤维化Ⅰ、Ⅲ型胶原基因表达的影响。方法 建立CCl4中毒性及人血白蛋白免疫损伤性 2种大鼠肝纤维化模型 ,在造模的同时给予不同剂量的重组人肝再生增强因子 (hALR)。在不同的时间点留取大鼠肝组织标本 ,提取总RNA ,用逆转录 聚合酶链反应 (RT PCR)测定Ⅰ、Ⅲ型胶原的基因表达水平。结果 在两种模型中hALR低剂量组大鼠肝组织I型胶原 (CCl4模型 2、4、6、8周分别为 1 .71± 0 .2 6、3 .42± 0 .67、3 .76± 0 .43、3 .3 3± 0 .69;人血白蛋白模型尾静脉攻击中、攻击后分别为 3 .86± 0 .66、2 .98± 0 .40 ) ,Ⅲ型胶原(CCl4模型 2、4、6、8周分别为 0 .97± 0 .1 6、2 .0 3± 0 .3 2、1 .86± 0 .3 3、1 .66± 0 .2 3 ;人血白蛋白模型尾静脉攻击中、攻击后分别为 2 .63± 0 .3 4、2 .0 2± 0 .3 2 )的基因表达水平在模型形成的不同阶段均明显低于模型组 ;高剂量hALR组大鼠肝组织Ⅰ、Ⅲ型胶原的基因表达水平均明显低于低剂量组。结论 重组人肝再生增强因子可能有抑制大鼠实验性肝纤维化Ⅰ、Ⅲ型胶原基因表达的作用。
Objective To investigate the influence of recombinant human augmenter of liver regeneration (hALR) on collagen Ⅰ and collagen Ⅲ gene expression in experimental liver fibrosis.Methods CCl 4 and albumin induced two kind of animal models of rat experimental liver fibrosis were established and different dosages of recombinant human augmenter of liver regeneration were given.Total RNA of liver tissues was isolated and collagen Ⅰ and collagen Ⅲ gene expression levels were detected by reverse transcription polymerase chain reaction (RT PCR).Results In both rat models of experimental liver fibrosis, collagen I gene expression levels were 1.71±0.26,3.42±0.67,3.76±0.43,3.33±0.69(CCl 4 model,2 weeks, 4 weeks,6 weeks,8 weeks),3.86±0.66,2.98±0.40 (albumin model before and after challenge);and collagen Ⅲ gene expression levels were 0.97±0.16,2.03±0.32,1.86±0.33,1.66±0.23 (CCl 4 model, 2 weeks, 4 weeks, 6 weeks,8 weeks),2.63±0.34,2.02±0.32 (albumin model before and after challenge). The gene expression levels in hALR treating group were significantly lower than those in the model group in different periods of the model (CCl 4 model, 2 weeks, 4 weeks, 6 weeks, 8 weeks, collagen Ⅰexpression levels were 3.26±0.43, 6.03±1.02, 6.46±0.93, 5.78± 0.53; collagen Ⅲ 1.34±0.26, 3.11±0.32, 2.63±0.34, 2.76±0.46. In the albumin model collagen Ⅰ8.03±1.17, 6.73±0.76 ; collagen Ⅲ 3.79±0.62, 3.03±0.43 before and after challenge respectively). Collagen I and collagen Ⅲ gene expression levels in high dose hALR treating group were significantly lower than those in low dose hALR treating group.Conclusion Recombinant human augmenter of liver regeneration might have effects of inhibiting gene expression of collagen Ⅰ and Ⅲ in experimental liver fibrosis.
