基因型和培养方式对大麦花粉植株高频率再生的影响 被引量:1

THE EFFECTS OF GENOTYPES AND CULTURE METHOD ON HIGH FREQUENCY OF ANDROGENESIS IN BARIEY

摘要 在基本培养基FHG上研究了基因型、培养方式、Ficoll 400及分化培养温度对大麦花粉植株再生频率的影响。结果,不同的基因型其花粉植株诱导频率不同。15个供试材料平均绿苗产量为每个花药产生1.46株,最低的是温Ⅰ,最高的是Igri的一个花培后代B631,分别为0.16和3.96株。平均愈伤组织诱导数为6.308块,绿苗分化率为23.24%,固体培养与液体培养比较,绿苗产量分别为每个花药2.31和4.15株。在液体培养基中补加Ficoll 400(100g/l),虽使愈伤组织诱导数和绿苗产量有所减少,但绿苗分化率及G/A值却有明显提高。分化培养时不同的培养温度其绿苗分化率也不同,20℃、15℃、10℃及25℃分别为38.27%、33.33%、26.67%和0%。 With FHG medium, the effects of genotypes, culture methods, Ficoll 400 and culture temperatures of dedifferentiation on regeneration frequency of green plant (RFGP) in barley were studied. The results are as follows: There were differences among different genotypes in regenerating pollen plants. Green plantlets were obtained in all fifteen materials tested. The yield of green plantlet (per anther) ranged from 0.16 for Wen I to 3.96 for B631-a pollen plant line of Igri . On the average, callus yield (per anther) was 6.308, the RFGP was 23.24%, the green plantlet yield reached 1.46. The green plantlet yield on liquid medium was 1.8 times higher than that on solid medium, reached 4.15 and 2.31, respectively. On liquid medium with Ficoll 400 (100 g /l), the callus yield and the green plantlet yield were reduced. But the RFGP and the G/A values were greatly increased. At different regeneration temperatures, there were greatly differences in the RFGP. The most suitable temperature for green plant regeneration was 20°C comparing with other three culture temperatures. The RFGP were 38.27% at 20°C , 33.33% at 15°C, 26.67% at 10°C and 0% at 25°C, respectively.
