环磷酰胺对小鼠骨代谢的影响及人参的防治作用探讨 被引量:5

摘要 目的 :观察环磷酰胺对小鼠骨代谢的影响并建立小鼠骨质疏松模型 ,同时观察人参对此模型的防治作用 ,并与葡萄糖酸钙进行比较。方法 :用环磷酰胺 ,造成小鼠以骨丢失为主的骨质疏松模型 ;并分别用人参和葡萄糖酸钙预防给药。各组实验结束后用原子吸收分光光度法和羟脯氨酸法测定该模型小鼠股骨的骨钙及骨羟脯氨酸含量。同时观察对照组和用药组的胸腺、肝、脾重量并进行比较。结果 :环磷酰胺导致小鼠胸腺萎缩 ,骨钙和骨羟脯氨酸含量等比例减少 ;人参可使环磷酰胺小鼠胸腺萎缩受到抑制 ;骨钙和骨羟脯氨酸含量增加。葡萄糖酸钙也可使环磷酰胺小鼠胸腺萎缩受到抑制 ;骨钙含量增加 ;但对骨羟脯氨酸含量没有显著影响 ,结论 :环磷酰胺可造成小鼠骨质疏松的病理模型 ,其特征为小鼠骨钙及骨羟脯氨酸含量等比例减少和胸腺萎缩。人参对此模型有明显的防治作用。葡萄糖酸钙也有阻止骨钙丢失和胸腺萎缩的作用 。 Objective:To observe the influence o f cyclophsphamide on bone metabolism and establish a model of mice osteoporosis. M eanwhile to determine the effects of Panax on the model and compare with Ca lcium gluconate.Methods:Cyclophsphamide was given to the m ice in order to induce bone loss.The femoral calcium contents of mice were assay ed by the method of Atomic Absorption spectrometry.The femoral hydroxyproline co ntents of mice were assayed by colorimetry.The thymus,liver and the spleen were weighed and compared.Results:Cyclophsphamide induced thym us shrinkage, the femoral calcium and the hydroxyproline contents decreased when compared to normal controled.Panax(5.0 g/kg·d) could reduce the shrinkage of t he thymus,increase the femoral calcium and hydroxyproline contents, when compare d to cyclophsphamide model group.Calcium gluconate(1.0 g/kg/d) could also reduc e the shrinkage of the thymus and increase the femoral calcium contents, but it couldn't affect the contents of femoral hydroxyproline.Conclusion:Cyclophsphamide can induce mice bone calcium contents loss and decrease bone hydroxyproline contents,with the shrinkage of the thymus of mice.Panax can prevent bone calcium and hydroxyproline contents loss and protect mice thymus fr om shrinkage.Calcium gluconate can also prevent bone calcium contents loss and p rotect mice thymus from shrinkage,but it can't prevent the loss of the femoral h ydroxyproline.
