摘要
目的 观察硝酸钴对大鼠胰岛A细胞的选择性损伤作用。方法 将 42只大鼠随机分为7组。对照组 6只肌注生理盐水 ,实验组肌注 1%硝酸钴盐液 5ml/kg体重 ,分别于注药后 1、2、3、4、6、10天取胰尾部 ,制成石蜡切片后进行免疫组织化学染色和体视学分析。结果 注钴后 2天胰岛A细胞开始出现损伤改变 (P <0 .0 5 ) ,3、4天损伤最为显著 (P <0 .0 0 1) ,6天后开始再生 ,10天后恢复正常 ,胰岛内B细胞、D细胞、PP细胞无明显变化 ,结论 用硝酸钴可选择性破坏大鼠胰岛A细胞 ,病变稳定 ,选择性高 。
Objective To observe the selective injury changes of Cobalt nitrate to islet A cells. Methods 42 rats were randomized into seven groups. In 6 rats of the control group, were injected with the solution of sodium chloride 36 rats of the experiment group were musclarly injected (5ml/kg) with the salts solution of cobalt nitrate. To draw the end of the pancreas 1,2,3,4,6,10 days after administration and to finish paraffin section for immunohistochemistry and stereology analysis. Results Islet A cells appear morphological injury after 2 days(P<0.05), become more significant after 3, 4 days(P<0.001), start to regenerate after 6 days and recover after 10 days. Injury wasn't seen in islet B cells. D cells and PP cells. Conclusion Islet A cells in rats were selective injured by cobalt nitrate and the pathological changes had a good stability and high selection rate.
