N-乙酰半胱氨酸对高糖环境中胰岛细胞的影响

摘要 目的探讨N-乙酰半胱氨酸(NAC)对高糖环境下胰岛细胞的影响。方法体外培养胰岛细胞INS-1,分为正常对照组,高糖组(葡萄糖25mmol/L),NAC作用组(浓度分别为50、100、200μmol/L),培养24h后,取上清测定胰岛素水平及谷胱甘肽过氧化物酶(GSH-Px)、丙二醛(MDA)含量。结果高糖可使胰岛素分泌量减少,GSH-Px浓度下降,MDA含量增加,NAC可以明显抑制高糖引起的上述变化,抑制作用随NAC浓度的增加而增强。结论抗氧化剂NAC对高糖导致的胰岛细胞损伤有一定的保护作用,并且呈浓度依赖性。 Objective To investigate the effect of N-acetylcysteine on islet cell cultured with high glucose. Methods The cultured islet cells(INS^l)were divided into 5 groups: normal eomrol group, high glucose group(glucose 25 mmol/L), 50/μmol/L NAC+PA group, 100/lmot/L NAC+PA group and 200 vtmol/L NAC+PA group. The level of insulin, GSH-Px and MDA in supernatant were measured when cultured for 24 h. Results High glucose had the effects of inhibiting insulin secretion of pancreatic islet cell, reducing the concentration of GSH-Px and enhancing the level of MDA. This change was inhibited by NAC with a dose-dependent manner. Conclusion NAC protected islet cell against injury induced by high glucose through inhibiting oxidation in dose-dependent manner.
