摘要
目的 探讨血管内皮生长因子 (VEGF)在结肠癌中的表达及其与微血管密度 (MVD)和p5 3之间的关系。方法 用免疫组化SABC法检测 6 8例结肠癌组织不同区域VEGF、MVD和 p5 3的阳性表达。结果 结肠癌区VEGF、MVD和 p5 3的表达明显高于癌旁区和正常区。结肠癌组织VEGF及p5 3的表达与肿瘤浸润深度、淋巴结转移、远处转移、血管侵犯及Dukes分期密切相关 ,而与组织学分型无关。p5 3(+ )或VEGF(+ )组MVD(34.6± 12 .2 ;31.2± 12 .6 )均显著高于p5 3(- )或VEGF(- )组(15 .0± 7.9;12 .7± 6 .3,P <0 .0 1) ;VEGF和 p5 3均为阳性时 ,MVD值最大 (36 .5± 11.9,P <0 .0 1)。MVD记数与VEGF表达明显相关 (P <0 .0 1) ,p5 3表达与VEGF表达和MVD记数均显著相关 (P <0 .0 1)。结论 在结肠癌血管生成过程中 ,可能存在 p5 3 VEGF调节旁路 ,p5 3基因在调控肿瘤血管形成方面起重要作用。
Objective To investigate the expression of vascular endothelial growth factor (VEGF) in colon cancer and its relation with the microvessel density (MVD) and p53 gene. Methods The expression of VEGF, p53 gene and MVD of different region of colon cancer were studied by immunohistochemical method from 68 by surgical resected specimens. Results The positive rates of VEGF, p53 and MVD in colon cancer were significantly higher than those in peritumor and normal tissue. The expression of VEGF and p53 positively correlated with depth of invasion, lymph node metastasis, distant metastasis, vessel invasion and Dukes' stage, but their relation to the histologic types was not significant. The MVD in p53(+) ( 34.6 ±12.2 ) and VEGF(+) (31.2±12.6) tumors was significantly higher than that in p53(-) and VEGF(-) tumors (15.0±7.9; 12.7±6.3, respectively, P <0.01), and MVD (36.5±11.9, P <0.01) in both p53(+) and VEGF(+) tumors was significantly higher than that in the other subgroups. A close correlation was present between MVD and VEGF expression ( P <0.01), and p53 expression correlated with both VEGF expression and MVD ( P <0.01). Conclusion It is possible that there is a p53 VEGF pathway which might regulate tumor angiogenesis in human colon cancer, p53 gene might play an important role in promoting tumor angiogenesis.
