摘要
目的 :探讨EB病毒感染细胞内Bcl 2水平与其对NK胞毒活性及对凋亡诱导因素敏感性之间的关系。方法 :应用反义寡聚脱氧核苷酸 ,抑制EB病毒转化B淋巴母细胞样细胞系 (EBV LCLs)胞内Bcl 2基因表达。然后观察NK细胞对其杀伤活性的改变以及其对致凋亡因素 (地塞米松、撤除生长因子 )敏感性的变化。结果 :EBV LCLs胞内Bcl 2表达水平与其对NK细胞杀伤的敏感性呈负相关 (P <0 0 1) ;与无血清培养条件下EBV LCLs细胞存活率呈正相关 (P <0 0 1) ;与EBV LCLs对地塞米松致凋亡作用的敏感性呈负相关 (P <0 0 1)。结论 :胞内Bcl 2水平可能是决定靶细胞对NK细胞杀伤活性敏感性的关键因素之一 ,NK细胞对EBV
Objective:To investigate the relationship between the level of Bcl 2 protein in EB virus infected cells and the sensitivity of this cells to NK activity and apoptosis inducing factors.Methods:Antisense oligodexynucleotides(ODNs) were used to modulate the expression of Bcl 2 gene in Epstein Barr virus transformed B lymphoblastoid cell lines(EBV LCLs);then the change of cytotoxicity of natural killer cells and the sensitivity of apoptosis inducing elements(taking out growth factor ?dexamethasone)targeting EBV LCLs,were investigated.Results:The level of Bcl 2 expression in EBV LCLs has negative relativity with the cytotoxicity of NK cells to EBV LCLs(P<0.01);positive relativity with survival rate of EBV LCLs in condition of no serum culture medium(P<0.01);and negative relativity with apoptosis sensitivity of the EBV LCLs to dexamethasone(P<0.01).Conclusion:The level of Bcl 2 in EBV LCLs may be the key element in determining NK's cytotoxicity targeting EBV LCLs;one mechanism of NK's cytotoxicity targeting EBV LCLs may be inducing cell apopotosis.
