摘要
目的研究温度对大鼠肝脏功能的影响,以确定大鼠肝脏的最适保存温度。方法采用单纯低温保存方法及建立大鼠原位肝移植模型研究保存温度对大鼠肝脏功能的影响。实验对0℃、4℃、8℃、12℃进行研究。保存时限为6,18及30h,保存18h 后行肝移植术。检测指标包括肝脏酶学、能量指标及组织形态学改变。结果①保存18,30h 的灌出液及移植后2h 血清中 AST、LDH 水平为4℃<0℃<8℃<12℃(P<0.01或P<0.05)。②保存18、30h 及移植后20min 的肝脏能量指标ATP、EC 为4℃>0℃>8℃>12℃(P<0.01或P<0.05)。③肝脏形态学示4℃组损害较其它组轻。结论 4℃对于大鼠肝脏是最适保存温度,它能够明显减轻大鼠肝脏的冷损伤程度。
Objective To investigate the storage temperature on liver function to determine the proximal storage temperature for liver preservation.Methods Cold liver preservation and orthotopic liver transplantation (OLT)were performed in rats.The liver was preserved for 6,18 and 30 hours in UW solution at specific tempera- tures(0℃,4℃,8℃ and 12℃).OLT was performed after the preservation for 18 hours.Liver function was de- termined by measurement of AST,LDH,ATP and EC.Meanwhile,the liver was observed with microscopy.Re- sults 1)The levels of AST and LDH were 4℃<0℃<8℃<12℃ at the 18th and 30th hour after the preserva- tion and at the 2nd hour after the transplantation(P<0.01 or 0.05).2) The levels of ATP and EC were 4℃> 0℃>8℃>12℃ at the 18th and 30th hour after preservation and at the 20th min after transplantation(P<0.01 or 0.05).3)The histological damage was slighter in 4℃ group than in other groups.Conclusions The tempera- ture of 4℃ is appropriate for hypothermic liver preservation in rats for relieve the cold ischemic injury.
