摘要
本文报道了从鼠细胞角蛋白基因组DNA文库中,经核酸分子杂交筛选的与鼠细胞角蛋白Endo B同源的阳性克隆No.4的亚克隆及核苷酸序列分析,又经微机辅助分析,证明No.4的461个核苷酸序列中,与Endo B cDNA同源性达93%,是与Endo B cDNA相关的假基因。
Four clones No.23, No. 4, No. 10, No.14 were screened from genomic DNA library encoding cytokeratin by hybridization. The DNA digested fragments with different orientation of clone No. 4 were subcloned into M_(13) phage vector, then sequenced by the dideoxynucleotide sequencing method. The nucleotide sequnce was analyzed and compared with Endo B structure DNA (clone No.23) and cDNA sequences using microcomputer. There aren't introns in No. 4. The overall homology is 93% between clone No. 4 and cDNA of Endo B for 461_(bp). However, one deletion in the 461_(bp) resulted in a serious frame shift, so No. 4 is a processed pseudogene corresponding to Endo B cDNA.
